ABSTRACT

Objective To prepare ebastine dispersible tablets by hot-melt dispersion method using hypromellose as a carrier.Methods Ebastine was heated to melt and dispersed in hot solution of hypromellose.The mixture was adsorbed by mannitol and then used to prepare dispersible tablets.The characteristics of hot-melt dispersion were analyzed by the methods of differential scanning calorimetry and X-ray powder diffraction.A comparison of dissolution curves between self-made dispersible tablets and original ebastine tablets was performed.Results The analysis of X-ray diffraction and differential scanning calorimetry showed that the ebastine in solid matrix remained the main crystalline characteristics.The dissolution of ebastine hot-melt dispersion was 95.07% in 15 min in vitro, which was significantly higher than that of micronized ebastine raw material.Moreover, the accumulative dissolution of ebastine dispersible tablets in 30 min was about 30% higher than the original drug.Conclusion The method of hot-melt dispersion using hypromellose as a carrier improved the ebastine dissolution significantly.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of modified bone morphogenetic protein-2 polylactic acid nanospheres (BMP-2-PLA-Ns) sustained-release system on rabbit mandibular defect repair.</p><p><b>METHODS</b>The polylactic acid s nanospheres (PLA-Ns) and BMP-2-PLA-Ns were prepared by ultrasonic emulsification after graft polymerization. Forty-five rabbits were randomly divided into 3 groups: Blank group, PLA-Ns gel group(control group), and BMP-2-PLA-Ns gel group (experimental group). The rabbit mandibular defect models were established. The defect area of control group was implanted with PLA-Ns gel, meanwhile, the experimental group was implanted with BMP-2-PLA-Ns gel, the blank group experienced no special handling. Rabbits were killed in 1, 2, 4 weeks after operation and the iconography, hematine eosin(HE) staining and PCNA immunohistochemistry were used to detect the reparative effect on rabbit mandible defects.</p><p><b>RESULTS</b>Image observation showed that bone defect repair in the experimental group was well and the shadow was not obvious. Better repair effect was seen compared with the control group and blank group. HE staining showed that the experimental group and the control group had a large number of neovascularization and secondary callus formation, callus in experimental group was obviously higher than that of control group and blank group. Immunohistochemical observation showed that the experimental group's PCNA positive chondrocytes were more than those in the control group and the blank group in the first 2 weeks; all groups of PCNA positive cells were rare in the fourth week, PCNA positive expression rate of the fourth week was lower than that of the first 2 weeks.</p><p><b>CONCLUSION</b>The modified BMP-2-PLA-Ns sustained-release system promotes mandibular defect repair obviously.</p>

ABSTRACT

Receptor activity modifying proteins (RAMPs) is a kind protein with transmembrane functionality.RAMPs can interact with TypeⅡ G protein coupled receptors (GPCRs) such as calcitonin gene-related peptide receptor (CGRPR),calcitonin receptor (CTR) and calcitonin receptor-like receptor (CRLR),to form a stable heterodimer expression on the cell membrane.Different RAMPs can combine with CRLR or CTR to produce different receptor phenotypes with ligand-specific affinity and thus can decide biological effect of the receptors.In addition,RAMPs can also interact with other GPCRs,which promise broader application in function regulation of G-proteincoupled receptor of RAMPs'.RAMPs' regulation of GPCRs depends on its molecular basis.Our studies of RAMPs provides a new perspective to further researches on GPCRs functionality and CGRP signal transduction.

ABSTRACT

[Objective]To provide a precise individualized design proposal for locating acetabular prosthesis by means of 3D reconstruction,reverse engineering software and rapid prototyping technology.[Method]3D CT scan pelvis image data were obtained from six patients,which displayed osteonecrosis of femoral head on one side.The data were transferred via a DICOM network into a computer workstation.A 3D model of hip was reconstructed using Amira 3.1 software,and saved in STL format.Then the 3D model was imported into Imageware 12.0 software.The normal hip joint centre of abnormal side was determined and the best orientation of the prosthesis(which was central axial ray of acetabula)was defined using reverse engineering.Locating template was designed according to the anatomic features of the acetabula and the central axial ray.Entity model was produced by rapid prototyping technology.The orientation of the prosthesis was located by matching the model to acetabular concavity at operation.[Result]By measuring postoperative standard hip anteroposterior film,the abduction angle of acetabular prosthesis was(46.00?2.49)? and the anteversion angle was(15.68?2.85)?.The expected result was obtained.[Conclusion]The navigation template designed and constructed preoperatively can provide precise location for acetabular prosthesis and avoid procedural errors at operation.
